Overview
FOX04-DRI is a D-retro-inverso peptide designed to disrupt the interaction between the FOXO4 transcription factor and p53 in senescent cells. By blocking this interaction, it is studied as a senolytic agent โ a compound that selectively induces apoptosis in senescent 'zombie' cells without affecting healthy cells.
Mechanism of Action
Senescent cells survive by exploiting the FOXO4โp53 axis to suppress apoptosis. FOX04-DRI is a cell-permeable peptide that competitively binds FOXO4, preventing it from sequestering p53 in the nucleus of senescent cells. This frees p53 to trigger apoptosis selectively in senescent cells, while normal cells remain unaffected.
